Product description


The UDG enzyme can catalyze the hydrolysis of the uracil base of the uracil-containing DNA strand and the N-glucoside bond of the glycophosphate skeleton to release free uracil. This product can act on dU-containing single-stranded or double-stranded DNA, and has no activity against RNA. The anti-contamination principle is that dUTP is incorporated into DNA instead of dTTP in the PCR reaction to form a PCR amplification product containing dU bases, and the UDG enzyme can selectively break the glycosidic bonds of dU bases in single-stranded and double-stranded DNA to degrade the PCR amplification products containing dU.

Unit definition


One unit (U) is defined as the amount of enzyme required to release 1 nmol of uracil from dU-containing single or double-stranded DNA within 5 minutes at 37°C.



Quality control


• Protein purity(SDS-PAGE): ≥95% .

• Endonuclease Activity: Incubation of 2 U of enzyme with 1 μg λ DNA for 16 hours at 37°C resulted in no detectable degradation of the DNA as determined by gel electrophoresis.

• Exonuclease Activity: Incubation of 2 U of enzyme with 1 μg λ-Hind III digest DNA for 16 hours at 37°C resulted in no detectable degradation of the DNA as determined by gel electrophoresis.

• Nickase Activity: Incubation of 2 U of enzyme with 1μg pBR322 for 16 hours at 37°C resulted in no detectable degradation of the DNA as determined by gel electrophoresis.

• RNase Activity: Incubation of 2 U of enzyme with 1.6μg MS2 RNA for 16 hours at 37 ℃ resulted in no detectable degradation of the DNA as determined by gel electrophoresis.



Precaution


1) This product is active in most PCR systems, and it is recommended to perform compatibility testing for the first self-use reaction system.

2) UDG enzyme should be stored at a low temperature during use.

3) For your health and safety, it is recommended to wear a lab coat and disposable gloves to perform the experiment.
