Background


Due to its high sensitivity, PCR technology is widely used in molecular experiments. However, during operation, it is prone to generating nucleic acid aerosol contamination. Even trace amounts of contamination can cause false-positive results, which is a major headache for researchers.

 

UDG (Uracil-DNA Glycosylase) can effectively hydrolyze the N-glycosidic bond between uracil bases and the sugar-phosphate backbone in single- or double-stranded DNA. When used in combination with dUTP, UDG specifically recognizes and degrades PCR products containing dU without affecting the template DNA. This enables efficient removal of aerosolized amplicon contamination and ensures the accuracy of PCR amplification results.


 

 

Currently, there are two main types of UDG enzymes available on the market: one derived from E. coli  or Bacillus subtilis  (standard UDG), and the other from psychrophilic marine bacteria (thermolabile UDG). Thermolabile UDG can be completely inactivated with a short incubation at relatively low temperatures.

 

Hzymes has launched a new product – Thermolabile UDG Enzyme – which can be fully inactivated at temperatures above 50°C. This effectively avoids the residual activity sometimes seen with standard UDG after heat inactivation, which could degrade dU-containing amplicons.

 


Product Overview


This product is highly temperature-sensitive and is completely inactivated at temperatures above 50°C. When combined with dUTP to form a contamination control system, it can be used in PCR, qPCR, RT-PCR, RT-qPCR, RT-LAMP, etc. A glycerol-free version of the thermolabile UDG enzyme is also available for use in lyophilized systems.

U-Product Digestion: In the presence of U-containing templates, Hzymes UDG demonstrates superior degradation ability compared to competing products A and B.


 


Heat Inactivation: Hzymes UDG is highly temperature-sensitive and becomes irreversibly inactivated after treatment at 50°C, showing better thermal inactivation performance than competitor A.


 


System Compatibility: Hzymes UDG does not interfere with amplification performance in either duplex or multiplex PCR systems.


 


Thermal Stability: In test systems, Hzymes UDG remains stable after accelerated incubation at 37°C for 10 days.
