Product Overview


Uric acid primarily reflects purine metabolism and renal excretory function, making it one of the core parameters in routine biochemical assays. Uricase (UA), also known as uric acid oxidase, is a key reagent component in uric acid detection kits. Due to its low water solubility, high concentrations of uric acid can precipitate in tissues, forming calculi and leading to gout. Uricase catalyzes the rapid oxidation of uric acid into allantoin, which is then eliminated from the body via urine through the renal tubules. Consequently, when used therapeutically in patients with hyperuricemia, uricase reduces serum uric acid levels, thereby alleviating and treating both urolithiasis and gout. Clinically, uricase is routinely employed to measure uric acid concentrations in serum and urine, assess renal dysfunction and hyperuricemia, screen for gout, and monitor long-term treatment efficacy. It also serves as an auxiliary enzyme to minimize uric acid-induced interference, enhancing the accuracy of other advanced assay kits. Under the action of uricase and oxygen, uric acid is converted into allantoin and H₂O₂; the latter, together with a chromogen, further reacts under peroxidase (POD) catalysis to produce a red quinone imine compound. The change in absorbance at a specific wavelength is directly proportional to the uric acid concentration in the sample. By comparing with uric acid calibration standards, the uric acid content in the sample can be quantified. Uricase (HH0901) is produced through directed engineering, microbial fermentation, column chromatographic purification, and lyophilization. Following formulation optimization, the resulting lyophilized product exhibits higher specific activity and markedly improved water solubility within UA assay systems. It demonstrates substrate specificity, selectively catalyzing only uric acid without cross-reactivity with purines or xanthines. The enzyme also displays superior thermal stability, with a half-life exceeding 1,700 minutes at 45°C-outperforming most commercial products. Its excellent short-term thermal tolerance at 60°C makes it ideally suited for enzymatic heat-treatment steps aimed at impurity removal and for drying dry-strip test formats. With a broad pH stability range of 5.5-10.0, optimal performance at pH8.5, and an ideal operating temperature of 37°C, it is well matched to standard UA assay platforms. Uricase (HH0901) has been scaled up for industrial production, available in multiple specifications and offering flexible customization. Its manufacturing process is robust, with minimal batch-to-batch variability, protein purity exceeding 90%, and extremely low levels of contaminating enzymes, ensuring reliable reagent performance. It is a standard raw material for liquid UA reagents used on global biochemical analyzers and for dry-strip test formats on POCT platforms, fully compliant with IVD raw material and reagent registration requirements.

Applications


Core applications: renal function-uric acid assay.


Uricase (HH0901), peroxidase (HH2801), and the Trinder chromogen are jointly used for the development and large-scale production of uricase-based assay reagents, and are equally compatible with POCT and rapid point-of-care uric acid dry test strips.


Secondary applications: Uricase serves as an auxiliary enzyme to mitigate uric acid interference.


Leveraging uricase's substrate specificity, the fact that uric acid consumes H₂O₂--the substrate in the Trinder reaction--and uric acid's strong absorbance in the 293/340nm UV region, uricase (HH0901) can be widely employed in Trinder-based assays--such as creatinine (SOX method), glucose (GOD method), cholesterol, triglycerides, and adenosine deaminase (XOD method)--as well as in NADH-based kinetic assays--for example, AST, ALT, urea, and homocysteine. By optimizing its formulation, this enzyme enhances the measurement accuracy of assay kits and improves their resistance to uric acid interference.



Reaction Principle


Uric acid levels can be measured at a wavelength of 290nm. One unit of enzyme activity is defined as the amount of enzyme required to oxidize 1μmol of uric acid per minute under conditions of 37°C and pH8.5.
