Product Overview


Cholesterol esterase (CE) is a recombinant enzyme with enhanced specificity for long-chain cholesterol esters containing C12, C18:1, and C18:2 fatty acid chains. It has been specifically developed for the differential detection of total cholesterol and high- and low-density lipoprotein cholesterol, and is compatible with the complete Randox quality-control and calibration system. Produced via high-density microbial fermentation followed by column chromatographic purification, it is manufactured on an industrial scale to ensure long-term supply stability. The lyophilized powder exhibits a specific activity of ≥90 U/mg, maintains stable activity over a pH range of 6.5–9.0, and withstands heat treatment at 60°C for 30 minutes. It demonstrates excellent compatibility with various nonionic surfactants and is well suited to specialized emulsification formulations for cholesterol assays. When combined with cholesterol oxidase (HH0504) and peroxidase (HH2801), it enables the construction of a complete enzymatic assay system for cholesterol measurement. The reagent exhibits outstanding resistance to interference from bilirubin, hyperlipidemia, and vitamin C, and retains its original performance even after accelerated testing at 37°C for 14 days. CE boasts high protein purity, with rigorously controlled levels of contaminating enzymes, ensuring no extraneous oxidase or esterase interference. It is ideally suited for largescale contract manufacturing of cholesterol reagents, as well as for developing standardized liquid and dry-strip IVD products for global manufacturers.

It remains almost completely inactive after 30 minutes at 60°C, demonstrating superior heat resistance compared to competing products.




Applications


Core Application – Lipid Profile Testing: The core raw material for cholesterol assay reagents, used in conjunction with cholesterol oxidase (CO, catalog no. HH0504) and peroxidase (HH2801), for the development and large-scale preparation of high- and low-density lipoprotein cholesterol as well as total cholesterol assays.



Reaction Principle



The amount of quinone imine produced in the reaction can be determined by measuring the absorbance at 555nm using a spectrophotometer. One unit of enzyme activity is defined as the amount of enzyme required to catalyze the formation of 1μmol of H2O2 per minute under conditions of pH6.0 and 37°C.
