α-L-fucosidase (AFU) is recombinantly expressed from thermophilic deep-sea microorganisms, serving as a highly pure, assay-specific calibrator and quality-control material for the diagnosis of liver diseases. It is paired with the specific substrate CNP-AFU (HH4701) to enable quantitative clinical measurement of AFU enzymatic activity. Compared with traditionally tissue-derived AFU, this product exhibits exceptional thermal stability: its optimal reaction temperature is 80°C, with a half-life of up to 214 minutes at 90°C; enzyme activity remains unchanged after prolonged incubation at 37°C. With a molecular weight of 55 kDa, it offers higher purity and lower impurity levels, maintains stable activity across a pH range of 7.0-10.0, and is compatible with a wide variety of buffer systems. Moreover, produced via large-scale microbial fermentation, it is not constrained by tissue or organ sources, allowing for reliable, high-volume supply. The enzyme solution can be stored sealed at -20°C for 12 months, retaining over 90% of its activity, and a lyophilized powder formulation is also available for even longer shelf life. It demonstrates excellent chemical compatibility across diverse buffer systems, and its linear calibration curve shows outstanding agreement, ensuring precise matching with AFU assays.
