Product Overview

Serum creatinine measurement is one of the routine clinical tests for renal function. As an important biochemical marker, it is used to diagnose and assess renal insufficiency, nephritis, and the efficacy of treatment for uremia caused by severe glomerular lesions, among other kidney diseases. Due to its poor stability and susceptibility to contamination, the alkaline picric acid method has been gradually phased out by major clinical laboratories; meanwhile, the creatine oxidase method has become the primary approach for clinical creatinine assay. Creatine oxidase (EC 1.5.3.1) catalyzes the oxidation of creatine to produce glycine, formaldehyde, and hydrogen peroxide, serving as the rate-limiting enzyme in the Trinder reaction coupled with the enzymatic creatinine assay. It exhibits resistance to interference from formaldehyde byproducts and effectively prevents absorbance drift. This product is compatible with standardized enzymatic creatinine colorimetric systems, featuring an enzyme specific activity of ≥15 U/mg, a protein purity of ≥90%, and tightly controlled levels of contaminating enzymes. It is optimized for the creatinine-specific pH range of 7.0-9.5 and demonstrates excellent long-term stability at 50°C. Compared with commonly available oxidase reagents on the market, this product offers a lower baseline signal and superior assay reproducibility, making it suitable for routine renal function testing in hospital central laboratories, as a reagent for fully automated biochemical analyzers, and for large-scale production of creatinine test strips. Its performance rivals that of leading international imported enzyme raw materials.

Applications

Core applications: Hydrolyzed creatine, used in conjunction with CAH (catalog no. HH0106), CR (catalog no. HH0203), and CAT (catalog no. HH2901) for the development and large-scale preparation of enzymatic creatinine assays.

Reaction Principle

The quinoneimine dye formed in the reaction can be detected at 555 nm. One unit of enzyme activity is defined as the amount of enzyme required to catalyze the production of 1 μmol of H2O2 per minute under conditions of pH 8.0 and 37°C.
