Product Overview

T7 RNA Polymerase (Thermostable) is suitable for in vitro transcription at higher temperature, using linear double-stranded DNA containing T7 promoter as template and NTPs as substrate to transcribe DNA sequences downstream of the promoter to obtain RNA. T7 RNA Polymerase (Thermostable) can also obtain biotin-, dye-, or radiolabeled RNAs using modified nucleosides as a substrate, and capped RNAs using cap structures or cap analogs as a substrate. The high-temperature reaction helps to reduce the formation of dsRNA byproducts and IVT product immunogenicity.

Key Features

Enhanced Thermostability and Catalytic Activity

Engineered through multiple rounds of directed iterative evolution, this thermostable T7 RNA Polymerase delivers simultaneous improvements in both thermostability and catalytic efficiency, enabling reliable performance in high-temperature in vitro transcription (IVT) systems.

Importantly, the enzyme also maintains excellent activity under conventional 37°C reaction conditions, allowing it to serve as a direct replacement for wild-type T7 RNA Polymerase without requiring process redesign.

Exceptional Performance at Elevated Temperatures

Compared with wild-type T7 RNA Polymerase, the enzyme exhibits a 270-fold longer half-life and 58-fold higher catalytic activity at 52°C, supporting more robust transcription under elevated-temperature conditions.

Improved Transcription of Challenging Templates

High-temperature IVT helps reduce the impact of RNA secondary structures, enabling more efficient transcription of templates with complex sequence features while maintaining high yield and transcript quality.

FAQ

Why choose Thermostable T7 RNA Polymerase over wild-type T7 RNA Polymerase?

Thermostable T7 RNA Polymerase is designed for high-temperature IVT workflows, enabling efficient transcription under elevated reaction conditions. Higher temperatures help minimize the impact of template secondary structures and may reduce dsRNA byproduct formation and the immunogenicity of IVT products.

What templates can be used with this enzyme?

The enzyme uses linear double-stranded DNA containing a T7 promoter sequence as the transcription template to synthesize RNA from the downstream DNA sequence.

Can this enzyme be used to synthesize modified or labeled RNA?

Yes. Thermostable T7 RNA Polymerase is compatible with a variety of modified substrates and can be used to generate:

Biotin-labeled RNA

Dye-labeled RNA

Radiolabeled RNA

Capped RNA using cap structures or cap analogs

This flexibility supports diverse research and therapeutic applications.

Does high-temperature IVT help reduce dsRNA formation?

Yes. Elevated-temperature transcription can help reduce the formation of dsRNA byproducts, which may contribute to lower innate immunogenicity of IVT products and reduce downstream purification burden.

What applications is this product suitable for?

This product is suitable for GMP-scale manufacturing and preclinical development of mRNA vaccines, gene therapies, circRNA applications, and other RNA-based therapeutics. It is particularly beneficial for templates with complex secondary structures and high-temperature IVT process development.

How should the product be stored and transported?

The product should be transported under temperatures below 0°C and stored at -25°C to -15°C to maintain optimal stability and activity.
