Product Overview

T7 RNA Polymerase (low dsRNA) is a DNA-dependent RNA polymerase from T7 phage, with highly specific 5'→3' RNA polymerase activity, and fewer dsRNA byproducts in the in vitro transcriptional product compared to wild-type T7 RNA polymerase. The production and quality management procedures for this product comply with GMP standards, ensuring traceability of the production process and all raw and auxiliary materials. The entire production process does not use ampicillin or any animal-derived raw materials. This product strictly controls residues of endonuclease, exonuclease, nickase, RNase, and endotoxins.

Key Features

Significant dsRNA Reduction Under Standard IVT Conditions

No changes to existing in vitro transcription (IVT) process parameters are required. Under the conventional 37°C reaction condition, this enzyme significantly reduces dsRNA byproducts, lowering the innate immunogenicity of mRNA at the source and substantially alleviating downstream purification burden.

Validated across a broad range of templates, dsRNA levels are typically reduced by more than 10-fold under standard 37°C conditions.

Broad Template Compatibility with Consistent Performance

Compatible with mRNA templates of varying sequences and lengths, the enzyme delivers robust dsRNA reduction while maintaining excellent transcription yield and RNA integrity. Its highly reproducible performance ensures outstanding batch-to-batch consistency for both process development and manufacturing.

Applications

GMP Manufacturing and Preclinical Development

Suitable for both GMP-scale production and preclinical research of mRNA vaccines, gene therapies, and other RNA-based therapeutics.

Personalized Cancer Vaccines

Supports the production of mRNA cancer vaccines requiring low innate immunogenicity and high transcript quality, helping improve therapeutic performance and tolerability.

Gene Editing Applications

Enables the generation of high-quality mRNA for gene editing platforms, where minimizing dsRNA impurities may contribute to improved safety and functionality.

Protein Replacement Therapy

Ideal for mRNA-based protein replacement therapies that demand enhanced safety profiles, robust manufacturing performance, and consistent product quality.

Performance Data

Reduces dsRNA content at 37℃

Validated against multiple templates, the M13 mutant has a significant advantage in reducing dsRNA content.

Lower immunogenicity

Template 1 and template 2 with co-transcription capping using M13 resulted in lower double-stranded content and transfection into RAW cells produced lower levels of the IFN-α and IL-6.

FAQ

What are the advantages of Low dsRNA T7 RNA Polymerase compared with wild-type T7 RNA Polymerase?

Compared with the wild-type enzyme, Low dsRNA T7 RNA Polymerase generates fewer dsRNA byproducts during in vitro transcription, helping reduce the innate immunogenicity of mRNA products and alleviate downstream purification burden.

What applications is this product suitable for?

This product is suitable for both GMP-scale manufacturing and preclinical development of mRNA vaccines, gene therapies, personalized cancer vaccines, gene editing applications, and other RNA-based therapeutics requiring low-dsRNA transcripts.

Is this product manufactured under GMP conditions?

Yes. The production and quality management procedures fully comply with GMP standards, ensuring complete traceability of the manufacturing process as well as all raw and auxiliary materials.

Are animal-derived materials or β-lactam antibiotics used during production?

No. The entire manufacturing process is animal-origin free and does not involve the use of ampicillin or other β-lactam antibiotics, minimizing the risk of exogenous contamination.

How is product quality controlled?

The product undergoes stringent testing for residual endonuclease, exonuclease, nickase, RNase, and endotoxin levels, ensuring high purity, reliable activity, and excellent batch-to-batch consistency.

How should the product be stored and transported?

The product should be transported under temperatures below 0°C and stored at -25°C to -15°C to maintain optimal stability and performance.
