Product Overview


D-Lactic dehydrogenase (DLDH) catalyzes the reversible conversion of pyruvate and NADH into D-Lactic acid and NAD+, exhibiting specificity for D-Lactic acid without cross-interference from L-lactic acid. It is a key enzymatic component in IVD reagents. This product is manufactured via high-density microbial fermentation followed by column chromatographic purification, produced on an industrial scale with a protein purity ≥90% and a specific activity of ≥420 U/mg in lyophilized form. Impurities are tightly controlled, batch-to-batch consistency is excellent, and long-term stable supply is ensured. Its unique advantages include a pyruvate affinity 2-3 orders of magnitude higher than that of competing products, broad pH stability from 5.0 to 9.0, and outstanding thermal stability. DLDH (HH1701) enhances the performance of HCY (cystathionine-based) and ALT/GPT assays, while also being well suited for assays targeting D-Lactic acid in bacterial infection typing, as well as metabolites such as pyruvate, ATP, glucose, and glycerol. HH1701 does not react with L-lactic acid in the blood, improving the accuracy of detection.



Extraordinary stability


Incubation at 50°C for 30 minutes maintains enzyme activity at over 90%. Incubation at 37°C for one week preserves dry-powder activity at over 90%. Storage at−20°C for one year also ensures that dry-powder activity remains above 90%. These characteristics provide robust stability for the development of diagnostic reagents targeting ALT, AST, and HCY.

Applications


D-LDH (HH1701), when used in conjunction with relevant enzymes, can be employed for the development and large-scale preparation of assay reagents targeting a variety of metabolites-including pyruvate, D-Lactic acid, HCY, ATP, ADP, glucose, creatinine, and glycerol-as well as enzymes such as alanine aminotransferase (ALT), pyruvate kinase (PK), and creatine kinase (CK).



Reaction Principle


The amount of NADH can be measured at 340nm using a spectrophotometer. One unit of enzyme activity is defined as the amount of enzyme required to oxidize 1μmol of NADH per minute under conditions of pH7.7 and 37°C.
