Product Overview

Catalase (CAT) catalyzes the decomposition of hydrogen peroxide into water and oxygen, specifically engineered as a matrix-compatible anti-interference agent for clinical two-step creatinine assay systems using dual reagents. Endogenous creatine is eliminated under the action of Creatine amidinohydrolase, Sarcosine oxidase and Catalase. When the activity of Catalase is inhibited under the action of NaN3, creatinine in the sample is converted into H2O2 under the action of Creatinine amidohydrolase, Creatine amidinohydrolase and Sarcosine oxidase. The generatedH2O2 can be detected using Trinder's reaction to quantitatively determine the creatinine in the sample.

With exceptionally high specific activity and controllable complete inactivation by sodium azide, it serves as a key functional enzyme for eliminating endogenous interferences. This product exhibits a specific activity of 13,000 U/mg, an ultra-wide pH stability range of 4.5–10.0, and excellent thermal stability below 65℃.

Its core advantage lies in efficiently removing endogenous peroxide-based interferences during the R1 pre-treatment step, while remaining fully inactivated by low concentrations of sodium azide during the R2 colorimetric reaction, with no residual enzymatic activity. It is primarily used for pre-processing in renal function serum diagnostic assays and is also suitable for applications in biopharmaceutical antioxidant formulations and the degradation of sterilization residues in cosmetics.

Key Features

High specific activity and high protein purity.

Good pH stability and chemical tolerance.

Long half-life at 70℃, with excellent thermal stability.

Low concentrations of sodium azide can completely inhibit CAT activity, enabling precise, timed inactivation.

Applications

Core Applications – IVD Clinical Diagnostics

① Creatinine-specific dual-reagent system:

The two-step creatinine assay uses R1 reagent to eliminate endogenous creatine interference.

② Other biochemical assays:

For tests such as uric acid, triglycerides, and total cholesterol, it is necessary to eliminate interference from endogenous hydrogen peroxide.

Extended Applications – Industry and Research

Antioxidant protection of proteins in biopharmaceutical processes, antioxidant treatment of cosmetic ingredients, and degradation of residual hydrogen peroxide in food-industry packaging and raw materials.

Reaction Principle

The amount of H2O2 consumed in the reaction can be determined by spectrophotometry at 240nm. One unit of enzyme activity is defined as the amount of enzyme required to catalyze the consumption of 1 μmol of H2O2 per minute under conditions of 37℃ and pH 7.0.

Performance Data

High Stability with Effective Elimination of Creatine Interference

After screening and optimization, HH2905 exhibits high specific activity and excellent thermal stability, maintaining substantial enzymatic activity even after 14 days of accelerated aging. A low dose of sodium azide can precisely inactivate CAT enzyme activity, completely eliminating endogenous creatine interference without affecting the primary creatinine reaction pathway.
