Technical Support

RNase R (Ribonuclease R) is a magnesium-dependent 3'→5' exoribonuclease derived from E. coli that digests essentially all linear RNA species while leaving circular RNA (circRNA), lariat RNA, and double-stranded RNA with short 3' overhangs (<7 nt) intact. Because it selectively removes linear RNA, RNase R is the standard tool for enriching circRNA and lariat RNA from total RNA — a critical step in both circRNA research and circRNA-based therapeutic manufacturing.

Product Description

Ribonuclease R (RNase R) from E. coli, is a magnesium-dependent 3’→5’ exoribonuclease that digests essentially all linear RNAs but does not digest lariat, circular RNA structures or double-stranded RNA with 3' overhangs less than 7 nucleotides. RNase R is commonly used in gene expression and alternative splicing studies to digest linear RNA for the enrichment of circular RNA (circRNAs) or lariat RNA. RNase R digests linear RNAs to enrich for circular RNAs used for intronic cDNA library construction from total RNA.

Applications

RNase R's linear-RNA - selective digestion makes it useful anywhere linear RNA needs to be removed without disturbing circular or branched RNA structures:

circRNA enrichment for research — depleting linear mRNA and rRNA from total RNA prior to circRNA-focused RNA-seq, allowing low-abundance circRNAs to be detected and quantified accurately.

circRNA vaccine and therapeutic manufacturing — as a purification step after in vitro circularization, RNase R removes residual unreacted linear RNA precursor and nicked/linearized byproducts, improving circRNA purity for downstream formulation.

Alternative splicing and gene expression studies — enriching lariat RNA intermediates (which retain a 2'-5' branch point) to study splicing intermediates that would otherwise be masked by the much larger pool of linear transcripts.

Intronic cDNA library construction — using RNase R to digest linear RNA from total RNA before reverse transcription, enabling libraries enriched for circular and lariat-derived introns.

Activity Unit Definition

One unit converts 1 μg of poly-r (A) into acid-soluble nucleotides in 10 minutes at 37 °C.

This definition anchors RNase R's activity to a standardized, reproducible substrate (poly-r(A), a synthetic linear RNA), so activity stated on the certificate of analysis is directly comparable between lots and consistent with the way RNase R activity is defined industry-wide.
