Product Overview

APS-5 chemiluminescent substrate is a new-generation chemiluminescent reagent for immunoassays. Formulated as an aqueous mixed solution based on APS-5 compound, this product achieves substantially improved performance via optimized key components including enhancers, stabilizers and preservatives. It generates chemiluminescence and rapidly emits photons upon catalysis by alkaline phosphatase (ALP). The maximum luminescence intensity is reached within approximately 2 minutes after sample addition with an extended plateau phase. Within the ALP concentration range of 10−4 to 10−8 U, the emitted photon count shows a linear positive correlation with ALP concentration in solution. Suitable for chemiluminescence immunoassay systems using ALP as the labeling enzyme, it features fast reaction rate, high sensitivity, superior stability and high signal-to-noise ratio.

Key Features

Rapid Signal Generation

Produces chemiluminescent signals rapidly following catalysis by alkaline phosphatase (ALP), supporting efficient assay workflows and automated detection systems.

Sensitive Detection

Capable of detecting ALP concentrations below 10¹ mol/mL and target proteins at levels as low as 5 pg, supporting applications requiring high analytical sensitivity.

Stable Luminescent Output

Provides a sustained luminescent signal with minimal impact from temperature fluctuations within the 25–35°C operating range, enabling reliable signal acquisition.

Broad Linear Range

Demonstrates a strong linear relationship between ALP concentrations ranging from 50 to 50,000 pg/mL and photon counts, supporting quantitative analysis across a wide dynamic range.

Ready-to-Use Formulation

Supplied as an optimized aqueous formulation containing enhancers, stabilizers, and preservatives, simplifying assay preparation and routine use.

Applications

ALP-Based Chemiluminescent Immunoassays

Suitable for chemiluminescent immunoassay systems utilizing alkaline phosphatase as the labeling enzyme.

Automated Diagnostic Platforms

Applicable to automated chemiluminescence analyzers requiring rapid signal generation and stable signal acquisition.

Immunodiagnostic Testing

Can be used in a variety of immunodiagnostic applications, including antigen and antibody detection assays.

Molecular Biology and Enzyme Activity Analysis

Suitable for molecular biology research and alkaline phosphatase activity studies involving chemiluminescent detection.

Performance Data

High Sensitivity with Low Background Signal

Compared with similar products from other manufacturers, Hzymes APS-5 luminescent reagent has obvious advantages such as lower background and higher signal-to-noise ratio.

Enhanced Thermal Stability and Photostability

After exposure to fluorescent light for 5 minutes, Hzymes APS-5 still maintains stable luminescence for 7 minutes.

After thermal acceleration at 37°C for 7 days, the luminescence value of APS-5 changes by 10%.

Ultra-Long On-board Stability for 28 Days

Average luminescence values (RUL) under multiple rounds of testing were measured at 2‒8°C, 25°C, and 30°C respectively. After 28 days of storage, the luminescence value decreased by ≤8%, demonstrating excellent storage stability of the product.

Excellent Repeatability and Batch-to-Batch Stability

Repeat testing and batch-to-batch variation tests were performed on Hzymes APS-5 chemiluminescent substrate solution. The CV < 3%, indicating that Hzymes APS-5 has excellent detection repeatability.

Test conditions: 5 μL enzymes + 150 μL substrate solution, enzyme activity 0.07 MU. All data were collected on a LUMI STATION 1800 instrument.
