GMP-Grade Murine RNase Inhibitor

RNase is quite universal in the environment, especially RNase A. RNase A is extremely stable and hard to be totally inactivated by autoclaving. RNases contamination is a main threat to the storage and stability of RNA samples. Common RNase inhibitors consist of chemical compound inhibitors and protein inhibitors. Among protein inhibitors, murine RNase inhibitor has improved resistance to oxidation compared to human/porcine RNase inhibitor, which means it is ideal for reactions where low DTT concentrations are required (e. g. , Real-time PCR, mRNA in vitro transcription).

In RNA-related experiments (cDNA/RT-PCR, mRNA in vitro transcription synthesis, polyribosome extraction), nuclease (RNase) contamination from pipette tips/tubes/other reagents or consumables can lead to the degradation of RNA. As a preventive measure, the recombinant RNase inhibitor will restrain RNase activity (RNase A/B/C) by forming a 1: 1 complex to ensure the stability and integrity of the RNA.

Thermal Stability and Freeze-thaw Stability Test of Haymes' RNase Inhibitor (Murine): The enzyme activity is not affected after 10 freeze-thaw cycles or when incubated at 55°C for 5 minutes.

Product Description

Murine RNase inhibitor is a recombinant murine RNase inhibitor expressed and purified from E. coli. It binds to RNase A, B or C in a 1: 1 ratio through non-covalent bonding, thereby inhibiting the activity of the three enzymes and protecting RNA from degradation. However, It is not effective against RNase 1, RNase T1, S1 Nuclease, RNase H or RNase from Aspergillus. Murine RNase inhibitor was tested by RT-PCR, RT-qPCR and IVT mRNA, and was compatible with various commercial Reverse transcriptases, DNA polymerases and RNA polymerases.
RNase inhibitor does not contain two cysteines that are highly sensitive to oxidation which causes inactivation of the inhibitor. That making it stable at low concentrations of DTT (less than 1 mM). This feature makes it suitable for use in reactions where high concentrations of DTT is adverse to the reaction (eg. , Real-time RT-PCR).
