Why Residual Enzyme Detection Matters in mRNA Manufacturing


During mRNA synthesis, several enzymes play essential roles:

DNase I is used to degrade unwanted DNA (plasmid or genomic DNA) contaminants. However, if not completely removed, residual DNase I can degrade RNA non-specifically, compromising mRNA integrity.
RNase Inhibitor (RNI) is employed to protect mRNA from degradation by ribonucleases. Residual RNI must be monitored to avoid interference with downstream applications.
Inorganic Pyrophosphatase (PPA1) enhances transcription efficiency by breaking down pyrophosphate (PPi), a byproduct that can inhibit RNA polymerase. Residual PPA1 could affect the stability of the final product.

Detection Principle


The assay kit operates on the double-antibody sandwich ELISA principle. Microplate wells are pre-coated with a capture antibody. Upon adding the sample and an enzyme-labeled detection antibody, incubation and washing steps form a capture antibody-antigen-enzyme-labeled antibody complex. After thorough washing, tetramethylbenzidine (TMB) substrate is added. TMB is catalyzed by peroxidase in the detection antibody to produce a blue color, which is subsequently converted to yellow by an acid stop solution. The intensity of the yellow coloration is directly proportional to the antigen concentration in the sample. The optical density (OD) is measured using a microplate reader, and the antigen concentration is calculated based on a standard curve.
