Introduction: Dengue Virus as a Global Health Challenge

Dengue virus (DENV) is a mosquito-borne flavivirus transmitted mainly by Aedes aegypti and Aedes albopictus. It includes four serotypes (DENV-1 to DENV-4), and secondary infections often increase the risk of severe disease.

Dengue has become one of the world’s fastest-growing infectious diseases, with the WHO estimating 100–400 million cases annually across more than 125 countries. Climate change, urbanization, and global travel continue to fuel its spread.

As researchers and public health institutions work to combat this crisis, the need for sensitive and specific molecular detection methods has never been greater. This is where Hzymes’ Dengue Virus Genotyping Nucleic Acid Detection Kit (PCR-Fluorescence Probing) provides critical value—offering reliable performance for dengue virus research and genotyping.

Product Overview: HMD8053 Dengue Virus Detection Kit

The Hzymes HMD8053 Dengue Virus Genotyping Nucleic Acid Detection Kit (PCR-Fluorescence Probing) is designed for the qualitative detection of dengue virus nucleic acids in serum and blood samples. This research-use-only kit enables laboratories to:

  • Detect dengue virus DNA with high accuracy

  • Differentiate between the four dengue virus serotypes (DENV I–IV)

  • Ensure robust performance through internal controls and anti-contamination safeguards

By enabling reliable nucleic acid detection, this kit contributes to epidemiological studies, molecular diagnostics research, and vaccine development strategies.

Key Features and Advantages

The Hzymes Dengue Virus Genotyping Kit is optimized for high performance and broad application.

  • High sensitivity: Limit of Detection (LOD) at 200 copies/mL

  • Precision and reproducibility: Ct coefficient of variation ≤ 5% across multiple runs

  • Specificity: No cross-reactivity with closely related viruses such as Yellow Fever, West Nile, Chikungunya, or Japanese Encephalitis Virus

  • Comprehensive genotyping: Detects all four DENV types (I–IV) in a single workflow

  • Built-in safeguards: Internal standard for process validation and contamination control system

These features make the kit a powerful research tool for dengue surveillance and outbreak monitoring.

Application Workflow: Step-by-Step Process

The Hzymes Dengue Virus Detection Kit is designed for ease of use and workflow efficiency in molecular biology laboratories.

  1. Nucleic Acid Extraction (Conducted in sample processing zone)

Manual and automatic processes are available for preparation of sample volume of 200 μL.

The DENV Positive Control, DENV Negative Control and the samples to be tested should be extracted simultaneously.

2. PCR Reaction Mixture Preparation (conducted in sample processing zone)

Thaw the DENV Reaction MIX 1 and DENV Reaction MIX 2 to room temperature, vortex, brief centrifuge, and aliquot 10 μL per PCR tube; transfer to the sample processing zone.

3. Sample Loading (conducted in sample processing zone)

Using filter tips, add 10 μL of test sample or controls to each tube, and cover the caps, briefly centrifuge, and transfer to the amplification or detection area, avoiding bubbles in the PCR tube.

4. PCR Amplification & Detection (amplification or detection area)

Place tubes in the real-time PCR instrument, record sample numbers and order, and set up PCR parameters as below (ABI 7500 example):

    • Threshold and baseline adjustments allow clear distinction between negative and positive results

This streamlined workflow enables laboratories to conduct fast and reliable dengue virus detection during critical research studies.

No.

Controls

Controls Standards

1

DENV Positive Control

The amplification curves of the FAM and ROX fluorescence channels in Reaction MIX 1 and Reaction MIX 2 exhibit a typical S-shape, and the detected Ct values are less than 40.

2

DENV Negative Control

The FAM and ROX fluorescence channels in Reaction MIX 1 and Reaction MIX 2 have no Ct value or a Ct value of 42.

All the above must be met in the test, otherwise the results are invalid.

Global Relevance: Supporting Dengue Virus Research Amid Outbreaks

The recent global surge in dengue infections emphasizes the importance of reliable molecular tools. With rising case numbers in Southeast Asia, Brazil, and even new cases in traditionally non-endemic regions such as Europe and the U.S., molecular surveillance and genotyping are critical.

The Hzymes Dengue Virus Genotyping Nucleic Acid Detection Kit provides researchers with:

  • A dependable tool for monitoring virus spread and mutations

  • Support for genotyping studies that inform vaccine and therapeutic development

  • Enhanced ability to study the epidemiology of dengue outbreaks

By delivering accuracy, sensitivity, and specificity, Hzymes is contributing to global dengue virus research and preparedness efforts.
